摘要:
目的:观察健骨颗粒冲剂对膝骨关节炎大鼠血清和关节液中肿瘤坏死因子-α及基质金属蛋白酶-3的影响,探讨健骨颗粒冲剂治疗膝骨关节炎的作用机制。方法:将48只Wistar大鼠适应性喂养1周后,随机抽取40只分为正常组、模型组、双醋瑞因组及健骨颗粒冲剂组,每组10只。分组结束后,将模型组、双醋瑞因组及健骨颗粒冲剂组大鼠制作膝骨关节炎动物模型。造模成功后,双醋瑞因组大鼠按0.009 g·kg-1以双醋瑞因溶液灌胃,健骨颗粒冲剂组大鼠按2.7 g·kg-1以健骨颗粒冲剂溶液灌胃,正常组和模型组给予相应剂量的生理盐水,每日1次,连续给药4周。药物干预结束后,采用酶联免疫吸附法检测各组大鼠血清和关节液中肿瘤坏死因子-α及基质金属蛋白酶-3的含量。结果:①血清肿瘤坏死因子-α含量。各组间含量比较,差异有统计学意义(F=10.056,P=0.008)。组间两两比较,模型组高于正常组、双醋瑞因组及健骨颗粒冲剂组(P=0.000,P=0.000,P=0.000); 其余各组间比较,差异均无统计学意义。②血清基质金属蛋白酶-3含量。各组间含量比较,差异有统计学意义(F=8.874,P=0.013)。组间两两比较,模型组高于正常组、双醋瑞因组及健骨颗粒冲剂组(P=0.000,P=0.000,P=0.000); 其余各组间比较,差异均无统计学意义。③关节液肿瘤坏死因子-α含量。各组间含量比较,差异有统计学意义(F=10.182,P=0.006)。组间两两比较,模型组高于正常组、双醋瑞因组及健骨颗粒冲剂组(P=0.000,P=0.000,P=0.000); 其余各组间比较,差异均无统计学意义。④关节液基质金属蛋白酶-3含量。各组间含量比较,差异有统计学意义(F=8.476,P=0.017)。组间两两比较,模型组高于正常组、双醋瑞因组及健骨颗粒冲剂组(P=0.000,P=0.000,P=0.000); 其余各组间比较,差异均无统计学意义。结论:健骨颗粒冲剂能降低膝骨关节炎大鼠血清及关节液中肿瘤坏死因子-α及基质金属蛋白酶-3的含量,从而减轻其对关节软骨基质的损害,直接或间接改善软骨基质代谢,促进软骨修复。
Abstract:
Objective:To observe the effect of JIANGU INSTANT GRANULES on tumor necrosis factor-α(TNF-α)and matrix metalloproteinase-3(MMP-3)in blood serum and synovial fluid for rats with knee osteoarthritis(KOA),and to explore the mechanism of JIANGU INSTANT GRANULES in the treatment of KOA.Methods:After 1 week adaptive feeding,40 rats were selected from 48 Wistar rats and randomly divided into normal group,model group,diacerein group and JIANGU INSTANT GRANULES group,10 cases in each group.After grouping,rats in model group,diacerein group and JIANGU INSTANT GRANULES group were built KOA animal models.After successful modeling,rats in diacerein group were administrated by gavage with diacerein solution(0.009 g·kg-1),rats in JIANGU INSTANT GRANULES group were administrated by gavage with JIANGU INSTANT GRANULES solution(2.7 g·kg-1),while rats in normal group and model group were administrated by gavage with corresponding doses of normal saline,once a day for 4 consecutive weeks.After drug treatment,TNF-α and MMP-3 contents in blood serum and synovial fluid of all the rats were measured through enzyme linked immunosorbent assay.Results:①TNF-α content in blood serum:there was statistical difference in the contents among the 4 groups(F=10.056,P=0.008).The content of model group was higher than that of normal group,diacerein group and JIANGU INSTANT GRANULES group(P=0.000,P=0.000,P=0.000); while there was no statistical difference in contents between any other groups.②MMP-3 content in blood serum:there was statistical difference in the contents among the 4 groups(F=8.874,P=0.013).The content of model group was higher than that of normal group,diacerein group and JIANGU INSTANT GRANULES group(P=0.000,P=0.000,P=0.000); while there was no statistical difference in contents between any other groups.③TNF-α content in synovial fluid:there was statistical difference in the contents among the 4 groups(F=10.182,P=0.006).The content of model group was higher than that of normal group,diacerein group and JIANGU INSTANT GRANULES group(P=0.000,P=0.000,P=0.000); while there was no statistical difference in contents between any other groups.④MMP-3 content in synovial fluid:there was statistical difference in the contents among the 4 groups(F=8.476,P=0.017).The content of model group was higher than that of normal group,diacerein group and JIANGU INSTANT GRANULES group(P=0.000,P=0.000,P=0.000); while there was no statistical difference in contents between any other groups.Conclusion:JIANGU INSTANT GRANULES can directly or indirectly improve cartilage matrix metabolism to promote cartilage repair through lowering TNF-α and MMP-3 contents in blood serum and synovial fluid for KOA rats to relieve their damage on articular cartilage matrix.
