摘要:
目的:探讨膝痹康对IL-1诱导的兔软骨细胞中MMP-3蛋白含量及MMP-3mRNA表达的影响。方法:把软骨细胞随机分为空白组(A组); 模型组(B组)及低、中、高剂量组(C、D、E组)并加以培养、干预。用酶联接免疫吸附剂测定(ELISA)法检测细胞MMP-3蛋白含量,再用两步法RT-PCR法检测MMP-3mRNA的表达水平。结果:空白对照组正常关节软骨细胞MMP-3蛋白含量及MMP-3mRNA表达量低于模型对照组,低、中、高剂量组组MMP-3蛋白含量及MMP-3mRNA表达量相对于模型组明显降低(P<0.01)。其中中剂量组MMP-3mRNA表达量最低(P<0.01)。结论:膝痹康含药血清可以抑制MMP-3的蛋白含量和MMP-3mRNA的表达水平,这可能是膝痹康中药延缓膝OA软骨退变,防止骨性关节炎的重要作用机制之一。
Abstract:
Objective:We study the machanism of the protection of Xibikang Prescription to chondrocytes induced by IL-1.Methods:The number of chondrocytes gained from wet cartilage, viability, multiplication and morphologic changes were observed. Chondrocytes were divided into different groups, XBK was added to subculture cells and its influence on protein levels of matrix metalloproteinase-3 was examined by ELISA method, and the influence on matrix metalloproteinase-3 mRNA expression were examined by RT-PCR method. Results:The levels of matrix metalloproteinase-3 protein and matrix metalloproteinase-3 mRNA expression.of the model control group was higher than that of the blank control group(P<0.01). The levels of high, middle and low dosage of Xibikang Prescription groups was lower than that of the model control group(P<0.05). and middle dosage of Xibikang Prescription groups was the most marked diffience with the model control group(P<0.01).Conclusion:Xibikang can effectively inhibit the levels of matrix metalloproteinase-3 protein and matrix metalloproteinase-3 mRNA expression.It plays a role in the mechanism of chondrocyte protection. It is effective to knee osteoarthritis patients.
