摘要:
目的:探讨桃红四物汤主次要有效成分的交互作用及其对血清VEGF浓度的影响。方法:从桃红四物汤中提取4类有效成分:总生物碱(A)、总苷(B)、总多糖(C)和总挥发油(D),按照前期实验所得结果(主要成分为A,次要为B、C、D),对清洁级小白鼠徒手定点折骨造模后进行实验。按照L8(27)正交设计方案分组,观察分析其对血清VEGF浓度的影响,探讨主次要成分之间是否存在交互作用。结果:骨折造模后断端局部出现VEGF蛋白高表达,提取物组表达量显著高于模型组(P<0.05); 动态观察结果,第2时相点(15 d)以内表达量逐渐升高,在第15天出现表达峰值,此后表达量开始下降; 各组间比较,1~8组、10组3个时相点表达量均明显高于9组; 在表达峰值(15 d)后VEGF表达量下降过程中,提取物及浓缩剂药物使表达量下降程度减缓,使其在第3时相点(25 d)断端骨痂组织内仍维持高量VEGF蛋白表达。正交实验表明,B药取水平1与水平2差异有统计学意义(P<0.05),C药和D药取水平1与水平2差异无统计学意义(P>0.05)。结论:有效成分总生物碱与总苷有交互作用,与总多糖和总挥发油无交互作用; 总苷、总多糖与总挥发油之间也无交互作用。
Abstract:
Objective:To explore the interactions of Taohongsiwu Decoction's(THSWD's)effective constituents and the influences on the dynamic change of VEGF protein expression of serum.Methods:Based on the former experiment,we extracted 4 effective constituents from THSWD:A(total alkaloids TA)who is the principal,B(total glycosides TG),C(total polysaccharide TPS)and D(volatile oil TVO)who is the secondary, executed experiment on the mus musculus albus after bare-handed fixed-point bone snaped, layed 8 groups according to the orthogonal design proposal, analyzed the VEGF protein expression of serum,which in order to study on whether there is interactions on Taohongsiwu Decoction's(THSWD's)principal and secondary or not.Results:There was higher VEGF expression after fracture molding than in the model(P<0.05). The result of dynamic viewing indicated that VEGF expression quantity was more in 1~8 group and in 10 group than in 9 group(P<0.05),and the quantity of VEGF expression increased at the 15th day and reached the peak value. Then the quantity of VEGF expression decreased but more slowly in the extraction group,which made the higher concentrations of expression were maintained in the region of bone stump.The orthogonal experiment indicated that there was obvious difference between B's level 1 and level 2(P<0.05).But there was no difference between C's or D's level 1 and level 2(P>0.05).Conclusion:there was interactions between TA and TC,but there was no interactions among TA,TPS and TVO,and so did among TC,TPS and TVO.
