摘要:
目的:观察血小板源性生长因子对末端病末端组织结构的影响,为血小板源性生长因子治疗末端病提供实验基础和理论依据。方法:对筛选后的大鼠随机分成3组(Ⅰ、Ⅱ、Ⅲ)组,其中Ⅰ组为血小板源性生长因子注射组,Ⅱ组为生理盐水注射组,Ⅲ组为对照组。将Ⅰ、Ⅱ组大鼠放入电刺激跳跃造模装置里,通交流电4 次·min-1,通电电压由小逐步加大,直到大鼠出现有效跳跃为止,共持续20 min,休息10 min,再持续20 min。每天训练1 次,每周6 次,周日休息,持续4周。实验过程中常规纯鼠料喂养。每周分别对血小板源性生长因子组和生理盐水组大鼠跟腱部位注射0.2 mL血小板源性生长因子和生理盐水,其注射时间均为大鼠休息日,对照组不做任何处理,造模4周后处死大鼠,进行取材,组织切片制作,HE染色。显微镜下观察每个样本跟腱末端、纤维软骨区、跟骨区、腱骨界面、腱围共5个区域病理变化。结果:血小板源性生长因子注射组大鼠跟腱末端区发生了轻微的病理性变化。生理盐水注射组大鼠跟腱末端区发生了明显的病理性变化。对照组大鼠跟腱末端区未发生明显的病理性变化。结论:注射血小板源性生长因子可以不同程度的有效地预防大鼠跟腱末端病的发生。
Abstract:
Objective:To observe the effects of platelet-derived growth factor on structure of tissue of terminal end disease to provide experimental and theoretical basis for its application in this disease.Methods:Screened rats were randomly divided into three groups,platelet derived growth factor injection group(Group Ⅰ),saline injection group(Group Ⅱ),control group(groupⅢ).Put Group Ⅰand Group Ⅱ into electrical stimulation jumping molding device,alternating current is 4 min-1,power voltage was gradually increased until the rats appeared effective jump with duration of 20 min followed with 10 min rest and 20 min alternating current again.Training was proceed once every day and 6 times per week with Sunday rest for 4 weeks.The experimental processed in conventional pure rat feed.Platelet derived growth factor group and saline group were applied with 0.2 ml each weekend on rat Achilles tendon while the control group without any treatment,modeling rats were sacrificed after 4 weeks,material,tissue section,HE staining were carried out.Microscopic observe each sample of Achilles tendon terminal,fibrocartilage zone,calcaneal tendon area,articular surface of bone,tendon in a total of 5 regional pathological changes.Results:Platelet derived growth factor injection group rat Achilles tendon terminal region has undergone minor pathological changes.Saline injection group rat Achilles tendon terminal region had obvious pathological changes.Control rats Achilles tendon terminal area without obvious pathological changes.Conclusion:Injection of platelet derived growth factor may vary in the degree of effective prevention of rat Achilles tendon terminal disease.
