摘要:
目的:研究骨碎补总黄酮对去卵巢大鼠腰椎松质骨骨形态计量学参数的影响。方法:将40只SD大鼠随机分为4组:假切除组(A组)、模型组(B组)、倍美力组(C组)、骨碎补组(D组),每组10只。麻醉后B、C、D组从背部入路切除双侧卵巢; A组仅切除腹部少许脂肪。造模后第2天开始,A、B组标准饲料常规饲养; C组在标准饲料常规喂养的同时,每天灌服倍美力悬剂2 mL(按人鼠体表面积换算,相当于倍美力0.017 mg); D组在标准饲料喂养的同时,每天灌服中药浓缩剂2 mL(按人鼠体表面积换算,相当于骨碎补总黄酮0.02 g)。6个月后取大鼠的L2椎体进行骨形态计量学检查。结果:4组大鼠骨体积分数、骨小梁厚度、骨小梁间距、骨小梁节点数及游离末端数比较,差异均有统计学意义(F=4.031,P=0.043; F=3.980,P=0.045; F=4.133,P=0.041; F=4.091,P=0.043; F=4.382,P=0.041)。同一指标组间比较:①B组骨体积分数低于A、C、D组(LSD-t=3.322,P=0.011; LSD-t=2.861,P=0.019; LSD-t=3.023,P=0.015); ②B组骨小梁厚度低于A、C、D组(LSD-t=3.360,P=0.011; LSD-t=2.925,P=0.019; LSD-t=3.066,P=0.017); ③B组骨小梁间距大于A、C、D组(LSD-t=3.417,P=0.011; LSD-t=2.981,P=0.019; LSD-t=3.108,P=0.013); ④B组骨小梁节点数少于A、C、D组(LSD-t=4.191,P=0.080; LSD-t=2.956,P=0.039; LSD-t=2.632,P=0.045); ⑤B组骨小梁游离末端数大于A、C、D组(LSD-t=4.021,P=0.008; LSD-t=2.237,P=0.048; LSD-t=2.510,P=0.038); ⑥C组与D组骨体积分数、骨小梁厚度、骨小梁间距、骨小梁节点数、骨小梁游离末端数比较,差异均无统计学意义(LSD-t=0.138,P=0.868; LSD-t=0.116,P=0.901; LSD-t=0.186,P=0.712; LSD-t=0.239,P=0.611; LSD-t=0.381,P=0.589)。结论:骨碎补总黄酮能通过增加骨量、改善骨三维结构,从而增强骨强度。
Abstract:
Objective:To study the effect of drynaria fortunei flavonoids on bone histomorphometry parameters of lumbar cancellous bone in ovariectomized rat.Methods:40 SD rats were randomly divided into 4 groups as sham-resection group(group A),model group(group B),premarin group(group C)and drynaria fortunei group(group D),10 cases in each group.Bilateral ovaries of rats in group B,group C and group D were removed from back approach after anesthesia,while the others in group A were administrated with resection of a little fat in belly only.From the 2nd day after molding,rats in group A and group B were fed on standard fodder normally,rats in group C were gavaged with 2mL premarin suspension agent(equivalent to 0.017mg premarin according to the conversion of human body surface area into rat body surface area )as well as fed on standard fodder every day,and rats in group D were gavaged with 2mL traditional Chinese medicine concentrate(equivalent to 0.02g drynaria fortunei flavonoids according to the conversion of human body surface area into rat body surface area )as well as fed on standard fodder every day.L2 centrums of rats were took for examination of bone morphometry after 6 months.Results:There was statistical difference in the following indexes as bone volume fraction,bone trabecular thickness,bone trabecular interval,number of bone trabecular nodes and number of bone trabecular free ends for the rats among the 4 groups(F=4.031,P=0.043; F=3.980,P=0.045; F=4.133,P=0.041; F=4.091,P=0.043; F=4.382,P=0.041).Comparison between groups for the same index:①Bone volume fraction of rats in group B were lower than those of rats in group A,group C and group D(LSD-t=3.322,P=0.011; LSD-t=2.861,P=0.019; LSD-t=3.023,P=0.015); ②Bone trabecula of rats in group B were thinner than those of rats in group A,group C and group D(LSD-t=3.360,P=0.011; LSD-t=2.925,P=0.019; LSD-t=3.066,P=0.017); ③Bone trabecular interval of rats in group B exceeded those of rats in group A,group C and group D(LSD-t=3.417,P=0.011; LSD-t=2.981,P=0.019; LSD-t=3.108,P=0.013); ④Bone trabecular nodes of rats in group B were less than those of rats in group A,group C and group D(LSD-t=4.191,P=0.080; LSD-t=2.956,P=0.039; LSD-t=2.632,P=0.045); ⑤Bone trabecular free ends of rats in group B were more than those of rats in group A,group C and group D(LSD-t=4.021,P=0.008; LSD-t=2.237,P=0.048; LSD-t=2.510,P=0.038); ⑥there was no statistical difference in bone volume fraction,bone trabecular thickness,bone trabecular interval,number of bone trabecular nodes and number of bone trabecular free ends of rats between group C and group D(LSD-t=0.138,P=0.868; LSD-t=0.116,P=0.901; LSD-t=0.186,P=0.712; LSD-t=0.239,P=0.611; LSD-t=0.381,P=0.589).Conclusion:Drynaria fortunei flavonoids can enhance bone strength through increasing bone mass and improving bone three-dimensional structure.
